Niacinamide, also known as vitamin B3 (VB3), is the bioactive form of niacin and is widely found in various plants and animals. It is also an important precursor to the cofactors NADH (nicotinamide adenine dinucleotide) and NADPH (nicotinamide adenine dinucleotide phosphate). Together with reduced NADH and NADPH, they act as coenzymes in more than 40 biochemical reactions and also function as antioxidants.

Niacinamide is a water-soluble substance with good solution and light stability. It exhibits optimal stability in a solution with a pH of 6.

1. Skin Whitening

Niacinamide can downregulate the transport of melanosomes from melanocytes to keratinocytes without inhibiting tyrosinase activity or cell proliferation, thus affecting skin pigmentation. It can also interfere with cell signaling pathways between keratinocytes and melanocytes, reducing melanin production. On the other hand, niacinamide can act on existing melanin, reducing its transfer to surface cells.

When used as a whitening ingredient, concentrations of 2% to 5% have been shown to be effective in treating melasma and hyperpigmentation caused by UV radiation. It is said that the effect is even better when combined with N-acetylglucosamine. Another perspective is that niacinamide also has anti-glycation properties, which can lighten the yellowing caused by protein glycation, helping to improve a sallow complexion and even a “yellowish” skin tone.

Studies have shown that using niacinamide (at a concentration of 5%) can reduce wrinkles, redness, yellowing, and age spots on aging facial skin. Therefore, it’s not surprising that Olay’s anti-aging product line contains a large amount of niacinamide. These effects may also be related to niacinamide’s ability to stimulate collagen production.

3. Improves Skin Barrier Function

Topical niacinamide can increase the levels of free fatty acids and ceramides in the skin, stimulate dermal microcirculation, and prevent moisture loss. It can also enhance protein synthesis (such as keratin), increase intracellular NADPH levels, and accelerate keratinocyte differentiation. Additionally, it can alleviate skin irritation caused by surfactants or solvents.

4. Moisturizing

The ability to improve skin barrier function, as mentioned above, means that niacinamide has moisturizing capabilities. A small study showed that topical application of 2% niacinamide was more effective than petrolatum (petrolatum) in reducing skin water loss and increasing hydration.

5. Oil Control and Pore Minimizing

Topical niacinamide (2%) can reduce the production of fatty acids and triglycerides in sebum, thus controlling sebum secretion. It is said to have a synergistic effect with sodium dehydroacetate. Reduced sebum secretion will also make pores appear smaller.

Treatment of Acne

It has been reported that gels containing 4% niacinamide are as effective as gels containing 1% clindamycin (a major topical prescription medication for treating acne) in treating acne vulgaris.
